Nucleotide Sequence Analysis of an Amylase Gene from Streptomyces albus KSM - 35

Abstract
The nucleotide sequence of the amylase gene (samy) from Streptomyces albus KSM-35 was determined using a series of deletion subclones. The samy gene contained an open reading frame of 1,380 nucleotides that encoded a protein of 460 amino acids with a deduced molecular weight of 49,639. The putative ribosomal binding site and the promoter site were found upstream of start codon. An inverted repeat located 27 bp downstream of the stop codon was likely to function as the transcription termination site. The samy gene has a high G+C content (70%), especially in the third position of codons. The 29 amino acid residues at the amino terminus has features similar to the signal sequences of other bacteria, which was composed of positively charged amino acid followed by a hydrophobic core. Sequence similarity between Samy and other amylases is quite limited. However, the four regions, which are involved in both the catalytic and calcium binding sites of various amylases, are highly conserved in Samy.
